The study was conducted to determine and compare the staining effect of C. longa, I. batatas and M. acuminata aqueous extracts for gonad examination of nile tilapia. Twelve nile tilapia fingerlings with approximately two and one-half to four-inch size were divided into four treatments with three replicates each. Treatment 1 was assigned as the positive control, which were stained with malachite green, while Treatments 2, 3 and 4 were stained with C. longa, I. batatas and M. acuminata extracts, respectively. The prepared slides of stained gonads were randomly distributed on the microscopes using completely randomized design (CRD)> Blind examination and evaluation by five faculty members was done on the stained unlabeled tissues under the microscopes. The tissues were evaluated according to five parameters: hue. saturation, contrast, clarity and overall appearance. A scoring scheme was followed, i.e. 1 to 2 - very poor; 3 to 4 - poor; 5 to 6 - average; 7 to 8 - good; and 9 to 10 - excellent. Data gathered were subjected to one way analysis of variance (ANOVA). The results suggested that C. longa, I. batatas and M. acuminata extracts are potential alternative stains for gonadal examinations of nile tilapia with M. acuminata as the most preferred.
