Formatted contents note | This study entitled "The Morphological, Molecular Identification and the Phylogeny of the Selected Varieties of Sweet Potato in Department of Agriculture CLIARC Tarlac" aimed to obtain the morphological characteristics of the species and the molecular identification of all the species (A-Fulfa Tarlac White (FTW), B-Orange Fleshed Sweet Potato (OFSP), C-Japonita, D-NSIC SP-32, E-Japanese Sweet Potato (SPJ), F-Taiwan Orange) using matK DNA markers; the relationship of the samples using Maximum Likelihood, Maximum parsimony and Neighbor Joining Tree. DNA Amplification was performed following the protocol of the angiosperms. The chloroplast maturase K gene (matK) is one of the most variable coding genes for angiosperm and has been suggested to be a "barcode" for land plants. Results show that the coding gene was amplified using matK DNA primer at 1570 bp. The species identity was recognized by their morphological and molecular characteristics. The purified sample were then sent to Macrogen, South Korea for DNA Sequencing. The raw DNA sequences were edited and assembled using Codon Code Aligner based on the peaks of the Consensus Sequences Chromatogram. The molecular identities of the samples were obtained based on BLAST Nucleotide Search results. They were identified as Ipomoea batata (AFulfa Tarlac White (FTW)99.43 % and 99.77 %, B-Orange Fleshed Sweet Potato (OFSP)99.54 %, C-Japonita 99.77%, D-=NSIC SP=-32 99.65%, E-Japanese Sweet Potato (SPJ)99.32 %, Fe Taiwan Orange99.43 %). |
